BaroFold provides client services in the area of protein refolding, aggregate characterization, formulation studies, and microbial expression, centered around its proprietary PreEMT technology. Client services supported by BaroFold include:
BaroFold conducts high-pressure refolding feasibility studies on all proteins, whether expressed from mammalian cells or E. coli. Having refolded approximately 100 proteins to date, BaroFold is highly knowledgeable in all redox, solution, and protein structural aspects of refolding. BaroFold conducts all feasibility studies in collaboration with the client, applying PreEMT technology and typically assessing yields in-house by employing analytical methods developed by the client. Applications include traditional refolds as well as pressure-treatment of final formulations to decrease aggregate content and reduce immunogenicity.
PROTEIN ANALYTICAL AND AGGREGATE CHARACTERIZATION
BaroFold conducts all protein biophysical and analytical methods for characterizing refolding and determining aggregate structures. We employ HPLC, CD, FTIR, 2D-UV spectroscopy, AUC, SDS-PAGE, bio-assays, and ELISA-based methods for protein analysis.
FORMULATION STABILITY AND IMMUNOGENICITY STUDIES
BaroFold conducts pressure and thermal-accelerated stability studies for clients to determine the stability of their protein formulations. Additionally, we are well-versed in designing studies to compare the immunogenicity of various formulations as a function of aggregate content.
BaroFold can conduct cytoplasmic expression of any client’s protein of interest, clone genes of interest in different expression vectors, and optimize expression.
